人参二醇组皂甙对游泳训练大鼠红细胞膜流动性的影响

摘    要:目的:探讨人参二醇组皂甙(PDS)防治长期耐力运动引起的红细胞损伤的作用。方法:稳态荧光偏振技术测定7周大运动量递增负荷游泳训练大鼠红细胞膜流动性,邻苯三酚-氯化硝基四氮唑蓝法测定红细胞超氧化物歧化酶活性,硫代巴比妥酸比色法测定血清丙二醛(MDA),扫描电镜观察红细胞形态。结果:运动·生理盐水组大鼠红细胞膜脂荧光偏振度(polatization,P值)显著高于安静对照组,反映红细胞膜流动性低于安静对照组(P<0.05);运动·生理盐水组的血清MDA水平显著高于安静对照组(P<0.05),而RBC-SOD活性显著低于安静对照组;运动·PDS组P值显著低于运动·生理盐水组,红细胞膜脂流动性高于运动·生理盐水组(P<0.05);运动·PDS组血清MDA水平显著低于运动·生理盐水组(P<0.01),RBC-SOD活性显著高于运动·生理盐水组(P<0.05);运动·生理盐水组的RBC计数、Hb、MCHC、Hb指数显著低于安静对照组(P<0.05);而运动·PDS组RBC计数、Hb、MCHC、Hb指数显著高于运动·生理盐水组(P<0.05);扫描电镜观察到运动·生理盐水组红细胞呈口形,棘形,嵴形,靶形等异常改变,而运动·PDS组红细胞形态基本正常。结论:PDS可降低运动训练后血清和红细胞脂质过氧化产物的形成,具有保护红细胞膜减少膜损伤的作用。

Abstract:AIM: To study effects of prolonged intensive endurance exercise and Panaxadiol saponins (PDS) on red blood cells (RBC) membrane fluidity in rats, as well as the mechanism. METHODS: By the membrane fluorence polarization, Pyrollol-NBT, TBA methods and Scanning Electron Microscope, RBC membrane fluidity,RBC-SOD and serum MDA were measured after 7 weeks prolonged intensive swimming. RESULTS: Fluorence polarization(P value) were significant higher and fluidity were lower( P <0 05), at the same time RBC-SOD activities lower and MDA contents higher in exercise-saline group than that of normal control group( P <0 05). P value were lower and fluidity were higher ( P <0 05), at the same time, RBC-SOD activities lower and MDA contents higher in exercise-PDS group than that of exercise- saline group( P <0 05). RBC count, Hb, MCHC and Hb index in exercise- saline group were significant lower than that of normal control group( P <0 05) and those parameter in exercise- PDS group were significant lower than that of exercise- saline group( P <0 05).Morphological study by scanning electron microscope showed that abnormal erythrocytes were found in exercise-saline group, while seldomly found in exercise-PDS group and normal control groups. CONCLUSION: PDS might reduce the lipid peroxidation in serum and RBC and has a good protective effect against RBC injury induced by prolonged intensive endurance exercise.
