灌注华蟾素对SD大鼠膀胱肿瘤抑制的实验观察

摘    要:目的:观察膀胱内灌注华蟾素(Cinobufagin,Cino)对SD大鼠膀胱肿瘤的影响。方法:75只SD雌性大鼠随机分成5组:空白对照组(A组)、MNU组(B组)、华蟾素处理组(C组)、阿霉素处理组(D组)、卡介苗处理组(E组),每组15只。所有SD雌性大鼠采用MNU膀胱灌注,每间隔1周灌注2 mg/0.2 ml 1次,共6次,诱导大鼠膀胱肿瘤模型。处理组分别予以华蟾素、阿霉素(ADM)、卡介苗(BCG)行膀胱内灌注。实验结束后1周处死大鼠,完整切除膀胱,膀胱肿瘤组织行HE染色,观察膀胱肿瘤病理类型,并计算各组的致癌率。结果:大鼠肿瘤组织HE染色,光镜观察A组大鼠14只大鼠均未见明显异常增生,B组13只大鼠发生膀胱移行细胞癌,C组6只大鼠发生膀胱移行细胞癌,D组7只大鼠发生膀胱移行细胞癌,E组5只大鼠发生膀胱移行细胞癌。结论:华蟾素能有效拮抗、抑制MNU诱导膀胱肿瘤的发生。

Abstract:Objective:To observe the effects by bladder injection of cinobufagin on bladder tumor in experimental SD rats.Methods:Seventy-five SD rats were randomized into groups of blank controls(group A),administration of N-methyl-N-nitrosourea(MNU),group B),treatment of cinobufatin(group C),adriamycin(ADM,group D) and bacillus calmette-guerin vaccine(BCG,group E)(n=15 for each).The animal models with bladder neoplasm were developed by bladder injection of MNU in dose of 2mg/0.2 ml,once every other week,for total 6 times.The treatment groups were managed with cinobufatin,ADM or BCG,respectively.One week after the final intervention,all rats were killed and complete bladder was removed.The tissues of bladder tumor were stained with hematoxylin and eosin(HE) for examining the pathological types and rate for carcinogenicity in each group.Results:Light microscopy showed no clear paraplasm in 14 rats in group A,yet transitional cell carcinoma of bladder in 13 rats in group B,6 in group C,7 in group D and 5 in group E,respectively.Conclusion:Cinobufagin may antagonize and even inhabit incidence of bladder caner induced by MNU.
