摘    要:采用组织化学-显微分光光度法和聚丙烯酰胺凝胶电泳法定量测定了工业品六氯环己烷染毒小鼠的精子、睾丸及附睾乳酸脱氢酶X同工酶(LDH-X)的含量,结果表现出明显下降并呈剂量-效应关系。提示工业品六氯环己烷能抑制小鼠生殖细胞LDH-X进而影响精子的能量代谢及运动能力。并对工业品六氯环己烷影响小鼠生殖细胞LDH-X的机理进行了讨论。

Abstract:LDH-X was found in different stages of spermatogenesis after primary spermatocytes and was located mainly in mitochondria of sperm. It was a specific mark enzyme related to the energy metabolism, development and genesis of germ cells. In this investigation,the influence of benzene hexachloride (BHC) on LDH-X of the germ cell in male mice was studied. Mice were administered by gastric intubation for 5 consecutive days. On the sixth day, they were killed by cervical dislocation, and one testis and epididymis were removed. Histochemical micro-spectrophotometry and polyacrylami-de gel electrophoresis were used to determine the activity of LDH-X quantitatively in mice testis, epididymis and sperm respectively. The results of histochemistry showed that the relative amount of special LDH-X in the neck-body of sperm from the three dosage groups was diminishedwith thin staining, which suggested the decline of enzymatic activity (p<0.05). Electrophoretic study showed that the activity of LDH-X in testicular homogenates of mice exposed to BHC was declined markedly (p<0.05). The activity of LDH-X of epididymis homogenate characterized mainly by mature sperm was parallel to that of testes, but the inhibition of LDH-X was more noticeable. BHC specifically influenced LDH-X activity of germ cell in mice, denoting that it could interfere with the energy metabolism of sperm, which was directly related to the decline of sperm count and motility. LDH-X is an early biochemical index showing chemical damage to germ cells.
