芦丁对雌性青春期大鼠乳腺发育及相关激素与受体水平的影响

摘    要:目的研究芦丁对雌性青春期大鼠内分泌激素水平、激素受体的表达以及乳腺组织形态学变化的影响,探讨芦丁促进乳腺发育的类雌激素样作用。方法 24只雌性青春期Wistar大鼠,随机分为3组,对照(Con):灌胃生理盐水2 ml/d;芦丁(Rut):灌服芦丁60 mg/(kg bw.d)一次;雌二醇(Est):肌注雌二醇60μg/(kg bw.w)。试验期2 w,结束时检测大鼠血浆和乳腺组织中雌激素(E2)、孕激素(P)、催乳素(PRL)、生长激素(GH)含量及其受体表达,并进行乳腺组织形态学观察。结果大鼠血浆与乳腺组织中E2、PRL水平Est组>Rut组>Con组(P<0.05);P水平Rut组>Con组(P<0.05),而与Est组差异不显著(P>0.05);GH水平Rut组与Con组与Est组差异不显著(P>0.05)。雌激素受体ER、孕激素受体PR、催乳素受体PRLR、生长激素受体GHR在乳腺组织中的表达量Rut组>Con组(P<0.05),Con组(P<0.05),与Est组差异不显著(P>0.05)。结论芦丁能够提高雌性青春期大鼠E2、P水平,促进PRL、GH分泌,上调乳腺组织中ER、PR、PRLR、GHR的表达量,促进乳腺发育。

Abstract:Objective To investigate the regulatory effect of rutin on the plasma level of endocrine hormones,the expression of hormonal receptors and the histomorphology of mammary glands in pubertal rats.Method Twenty four intact virgin rats were randomly assigned to three groups: 1) control group(Con):i.g.2 ml normal saline per mouse per day;2) rutin group(Rut): i.g.60 mg rutin/kg bw per day;and 3) estradiol group(Est):i.m.60 ?g estradiol/kg bw weekly.The trial lasted for 2 w.The concentration of estrogen(E2),progesterone(P),prolactin(PRL) and growth hormone(GH) in plasma and gland tissues,and the expression of the receptors of these hormones(ER,PR,PRLR,GHR) in gland tissues were measured.The histomorphology of mammary tissues was also observed.Results The concentration of E2 and PRL in plasma and mammary tissues was ranked as Est>Rut>Con(P<0.05).and the concentration of P in plasma and mammary tissues was Rut>Con(P<0.05).The concentration of GH in plasma and gland tissues was insignificantly different among Rut,Con,and Est groups(P>0.05).The expression of ER,PR,PRLR and GHR in mammary tissues was Rut>Cont(P<0.05) but RutCont(P<0.05) but insignificantly different to Est group(P>0.05).Conclusion Rutin improved the plasma concentration of E2 and P in intact virgin rats,increased PRL and GH release,and up-regulated the expression of ER,PR,PRLE and GHR,which accelerated the growth of mammary gland.
