摘    要:Objective To examine the role of 5-HTEc receptor in pulmonary arterial smooth muscle cells (PASMCs) proliferation and contraction induced by 5-HT. Methods Small interfering RNA (siRNA) was used to knock down the expression of 5-HTEcR. We used MTT assay and cell cycle analysis to demonstrate the proliferation of PASMCs. The length of PASMCs was measured to show the cells contractile function. Results Administrated of 5-HTEcR-specific siRNA, 5-HTEc receptor expression in PASMCs was significantly suppressed. MTT assay and cell cycle analysis showed that 5-HT-induced cell growth in PASMCs was attenuated. Contraction of PASMCs induced by 5-HT was also obviously reduced. Conclusion 5-HTEc receptor mediates the proliferation and contraction of PASMCs induced by 5-HT.
