α-内收蛋白Gly460Trp基因多态性与复方缬沙坦降压疗效的关系

目的:探讨内收蛋白α亚单位(α-adducin)Gly460Trp基因多态性对复方缬沙坦降压疗效的影响.方法:采用突变分离聚合酶链反应(MS-PCR)检测α-adducin基因Gly460Trp多态性.以96例轻中度原发性高血压患者为研究对象,给予复方缬沙坦治疗8周进行自身对照研究;同时测定身高、体重、生化检查、血压等临床资料.结果:EH患者存在α-adducin基因Gly460Trp多态性,复方缬沙坦对EH患者α-adducin TT基因型者降低收缩压的作用强于GG GT基因型者(P＜0.05).结论:EH患者α-adducin基因460Trp突变纯合子者服用复方缬沙坦后降低收缩压可获得较好的疗效.

Association of Gly460Trp Polymorphism of alpha - adducin Gene and the Effective of Valsartan/Hydrochlorothiazide

Objective: To investigate the association of the Gly460Trp polymorphism of the alpha-adducin gene with the antihyperten sive effects of valsartan/hydrochlorothiazide in hyper-tensive patients. Methods: The Gly460Trp polymorphism of alpha - adducin in 96 hypertensive patients was determined by mutagenically separated PCR (MS - PCR). They were treated with valsartan/hydrochlorothiazide for 8 weeks and were examined by physical examination and biochemical analyses. Results: Adducin Gly460Trp existed in hypertensive patients. Hypertension treating with valsartan/ hydrochlorothiazide for 8 weeks, patients with the adducin gene homozygous for T allele showed a greater reduction in systolic blood pressure than non - Trp/Trp (GG + GT) . Conclusion: Hypertensive patients with the adducin gene homozygous for T allele showed a greater reduction in systolic blood pressure for valsartan/hydrochlorothiazide treatment.
