摘    要:探讨同型半胱氨酸 (Hcy)诱导人脐带静脉内皮细胞株———ECV30 4细胞氧化损伤的机制。用含不同剂量Hcy(0 0 5— 2 0 0mmol L)的培养基培养细胞 1 2小时以后 ,以分光光度比色法分别测定细胞内丙二醛含量和超氧化物歧化酶、谷胱甘肽过氧化物酶、过氧化氢酶活力的改变。以荧光显色法测定细胞内活性氧产生的改变。结果表明小剂量 (0 0 5~ 0 1 0mmol L)Hcy可明显的促进氧化损伤 ,促使细胞内MDA含量增加和细胞内活性氧的产生 ,并显著降低细胞内除过氧化氢酶以外的其它抗氧化酶 (超氧化物歧化酶和谷胱甘肽过氧化物酶 )的活性。结论认为Hcy可能通过诱导细胞氧化损伤而损害血管内皮细胞

Abstract:The mechanism of homocysteine-induced oxidative injury on ECV304 cell line, a spontaneously transformed immortal human endothelial cell line, was studied. ECV304 cells were incubated with different levels of Hcy (0.05-2.00 mmol/L) for 12 hours. The activity of major anti-oxidative enzymes and the generation of reactive oxidative species (ROS) were measured by chromatometry and fluorochromy. The results showed that Hcy could induce obvious oxidative injury on ECV304 even at low level (0.05-0.10 mmol/L). The level of MDA was increased and the active oxygen was generated in the cells. The activities of anti-oxidative enzymes (SOD and GSH-Px) except the activity of CAT were decreased in the cells. It was concluded that Hcy may induce oxidative injury on EVC304 cell line through increasing the levels of ROS and inhibiting the activities of anti-oxidative enzymes.
