摘    要:目的:通过噬菌体特性观察,为了解噬菌体吸附特异性及溶苗机理的分子生物学机制奠定物质基础.方法:利用标准苗株从污水中分离特异性噬菌斑,纯化增殖后得到其相应噬苗体.经过高滴度噬菌体与其非特异性宿主菌的混合培养,筛选出宿主谱特异性发生突变的具有宽噬性的噬菌体.结果:从污水中分离出12种特异性噬菌体,发现了可裂解同一菌属的不同菌种的宽噬噬菌体,改变了噬菌体专-宿主谱这一特性.结论:针对同一菌属中不同菌种的宽噬噬菌体具有较高的稳定性及裂菌滴度,为探索噬菌体寄生的基因控制及特异吸附的分子生物学机制奠定了基础,将人工改造噬菌体,使之成为新型的杀菌生物制剂成为可能。

Abstract:AIM: To understand the specific adsorption and bacteriolysis mechanism by observing the biological characteristics of the bacteriophage. METHODS: Use standard strain to separate specific phage from sewage and then to screen the mutation phage with broad host range. RESULTS: We had separated 12 kinds of phages from sewage and found the broad host range phage that could lysis different bacteria in the same genus. CONCLUSION: The broad host range phages have high stability and bacteriolysis titer, which can be used to study the gene control of phage parasitism and specific adsorption.
