摘 要: | 目的:探讨电针百会、太阳穴对帕金森大鼠多巴胺能神经元凋亡的作用机制。方法:采用单侧纹状体双靶点注射6-OHDA建立大鼠帕金森病模型。实验分假手术组、模型组、阳性药组(美多巴组)和电针组,分组给予相应处理4周。分别应用免疫组化方法检测黑质半胱氨酸天冬氨酸蛋白酶3(Caspase-3)的表达情况和原位末端标记法(TUNEL)检测细胞凋亡水平。结果:模型组大鼠脑黑质Caspase-3表达明显增多,多巴胺神经元细胞凋亡显著,电针百会、太阳穴可使Caspase-3表达下调,使多巴胺神经元细胞凋亡程度减轻。结论:电针百会、太阳穴治疗帕金森病的作用机制可能是通过减轻Caspase-3的表达,抑制细胞凋亡减少多巴胺神经元的丢失来实现的。

Effects of Electro - acupuncture on Baihui and Taiyang Points on Caspase - 3 Expression and Apoptosis of Substantia Nigra Dopaminergic Neurons of Rats with Parkinson Disease |

Abstract: | Objective: To explore the mechanism of protective effect of electro - acupuncture on Baihui and Taiyang points on dopaminergic neurons of rats with Parkinson Disease. Methods: Models with PD were made by two targets injecting 6 - OHDA in one side of striatum. Erperiment was divided into sham - operation, model, positive and electro - acupurcture groups, each group was given the cornesponding treatment for 4 weeks. The expression of Caspase - 3 in the substantia nigra was determined by immunohistochemistry method, the apoptosis of neurons was examined by TUNEL method. Results: The Caspase - 3 expression in model group increased ob- viously and the apoptosis level of neurons was obviously increased, while electro - acupuncture on Baihui and Taiyang points can both down - regulate the Caspase - 3 expression and alleviate the apoptosis level. Conclu- sion: Electro- acupuncture on Baihui and Taiyang points can effectively treat PD, decreasing the Caspase -3 expression and reducing the loss of dopaminergic neurons maybe one of the mechanism. |
