电针合谷穴对痛经大鼠扭体行为、PGE_2、PGF_(2α)、β-EP含量的影响

摘    要:目的:初步研究电针合谷穴对痛经大鼠的镇痛效应及机制。方法:将30只处于动情间期的SD雌性大鼠随机分为3组:正常组、模型组、电针合谷组。采用苯甲酸雌二醇和缩宫素联合制备痛经大鼠模型,观察电针合谷穴对痛经大鼠扭体行为、前列腺素F2α(PGF2α)、前列腺素E2(PGE2)、β-内啡肽(β-EP)含量的影响。结果:与模型组比较,电针合谷穴组大鼠扭体潜伏期明显延长,扭体次数、总分均显著性减少(均P0.01);子宫PGE2含量无显著性差异(P0.05),PGF2α含量及PGF2α/PGE2比值均显著性下降(均P0.01);血浆、子宫、下丘脑β-EP含量均无显著性差异(均P0.05),垂体β-EP显著性升高(P0.01)。结论:电针合谷穴可以通过降低大鼠子宫PGF2α含量和PGF2α/PGE2比值、升高垂体β-EP含量达到止痛的作用,其镇痛效应机制可能是通过提高中枢β-EP的含量达到的。

Effect of Elecrt o-acupuncture at Hegu Point on Writhing Behavior,PGE2, PGF2αandβ-EP Levels in Dysmenorrheal Rats

Abstract:Objective:To preliminarily study the analgesic effects of electroacupuncture ( EA ) at Hegu (LI4)point in dysmenorrhea rats and explore the mechanism .Methods:30 SD female rats aged three-month-old in the estrum were randomly divided into normal group, model group and EA group. Rats were given estradiol benzoate combined with oxytocin to prepare the models of dysmenorrhea and normal group was given the same dose of saline , EA group was given EA .The effect of EA at Hegu on the writhing behavior , PGE2、PGF2α、β-EP levels in dysmenorrheal rats were observed .Results:To compared with model group , the rats'writhing incuba-tion period in EA at LI4 group was obviously prolonged ,rats'writhing number and scores were decreased signifi-cantly(all P〈0.01);there was no significant difference between the two groups on uterus PGE 2 level (P〉0. 05);PGF2αlevel and PGF2α/PGE2 ratio were reduced significantly in EA group (all P〈0.01);β-EP level in plasma,uterus and hypothalamus had no significant difference (all P〉0.05), in pituitary that was significantly increased(P 〈0.01).Conclusions:Electro-acupuncture at Hegu point can reduce PGF 2αlevels and PGF2α/PGE2 ratio in the rats'uterus,increae β-EP level to achieve analgesic effect .
