超声心动图评价干细胞移植对心肌梗死大鼠左心室功能的影响

摘    要:目的采用二维超声心动图和组织多普勒超声心动图评价骨髓干细胞(bonemarrow-drivedmesenchymalstemcells,MSCs)移植前后大鼠的心脏形态和左心功能,为干细胞移植治疗心肌梗死的临床应用提供参考数据。方法40只Wistar大鼠随机分为对照组和移植组。应用液氮冷冻法制作心肌梗死模型后,移植组大鼠心肌内注射经5-氮胞苷诱导后的MSCs。手术前、术后1周、术后1个月分别应用二维超声心动图和组织多普勒显像测定两组大鼠的心脏功能。结果术后1个月移植组和对照组大鼠分别存活13只和15只。数据分析表明:移植组术后1个月,大鼠心脏功能与术后1周时相比明显改善(P<0.05),而对照组无改善。结论诱导后的MSCs移植到心肌梗死区及其周围可防止心功能恶化,改善心脏功能。MSCs移植可能成为治疗心肌梗死的一种切实有效的方法。

Abstract:Objective To observe the rat cardiac function change before and after bone marrow stem cells(MSCs) transplantation by two dimensional echocardiography and tissue Doppler echocardiography. Methods Forty rats were divided into transplanted group and control group randomly. After the model of myocardial infarction of the rats were established, MSCs were transplanted into the infarcted sites of the transplanted group. The equal quantities of culture fluid were injected into the myocardium of the control group. Before and 1 week, 1 month after operation, the heart function was detected by echocardiography. Results Thirteen rats of transplanted group and 15 rats of control group survived 1 month after operation. The heart function improved in transplanted group after operation while the heart function of control group deteriorated. Conclusions To transplant cultured MSCs into infracted rat hearts can improve heart function. The transplantation of exogenous MSCs for treatment of myocardial infarction may be feasible and effective in rat heart.
