摘    要:目的研究Gravin在血管内皮生长因子(VEGF)诱导人脐静脉内皮细胞(HUVECs)迁移和体外“小管样结构”(TLS)形成中的作用。方法用VEGF诱导HUVEC,通过Western blot和细胞免疫荧光分析Gravin在细胞内分布的变化;用Gǒ6983【蛋白激酶C(PKC)抑制剂】和VEGF共处理内皮细胞,观察对Gravin分布的影响,观察其对VEGF诱导HUVECs迁移和TLS形成的影响。结果VEGF呈时间和剂量依赖性诱导Gravin磷酸化,并向核周和细胞边缘重分布;以上效应能被0.5μmol/LGǒ6983抑制;Gǒ6983同时抑制VEGF诱导HUVEC迁移和TLS形成。结论PKC-Gravin信号通路参与VEGF诱导的内皮细胞迁移和血管形成。

Abstract:Objective To observe the role of Gravin structure (TLS) of human umbilical vein endothelial cells on the migration and formation of tubule-like (HUVEC) in vitro stimulated by vascular endothelial growth factors (VEGF). Methods Cultured HUVEC were stimulated by different concentrations of VEGF. Gravin expression was detected by Western blot and immunofluorescence technology. Cultured HUVECs were co-treated with Gǒ983 (PKC inhibitor) and VEGF, Gravin expression was detected, HUVECs migration and the TLS formation were detected. Results VEGF stimulated Gravin phosphoryla- tion in time and concentration dependent manner and relocation from the diffuse distribution to periphery and nuclear sites ; Gǒ983 inhibited the former effects of VEGF on Gravin. In addition, Gǒ66983 inhibited migration and TLS formation of HUVEC stimulated by VEGF. Conclusion PKC-Gravin signaling pathway may play an important role in HUVECs migration and TLS formation stimulated by VEGFs.
