人胃癌细胞和胃癌组织中胃肠富集Kruppel样因子和肠道富集Kruppel样因子的表达及其临床意义

摘    要:目的研究人胃癌细胞株和胃癌组织中胃肠富集Kruppel样因子(GKLF)、肠道富集Kruppel样因子(IKLF)的表达及其临床意义。方法采用免疫细胞化学法检测人正常胃上皮细胞株GES-1及胃癌细胞株MKN28、SGC7901、BGC823、HGC27中GKLF、IKLF的表达;采用免疫组织化学法检测96例胃癌组织及其远端正常胃组织中GKLF、IKLF的表达。结果GKLF在人正常胃上皮细胞株内阳性表达,在胃癌细胞株内无表达;IKLF在人正常胃上皮细胞株内无表达,在胃癌细胞株内阳性表达。胃癌组织中GKLF阳性率为20.8%,显著低于远端正常胃组织(64.6%,P〈0.01),胃癌组织中IKLF阳性率为71.9%,显著高于远端正常胃组织(21.9%,P〈0.01)。GKLF、IKLF表达与胃癌患者的年龄、性别、分化程度、Lauren分型无关(P〉0.05),但与淋巴结是否有转移及TNM分期显著相关(P〈0.01)。结论GKLF、IKLF在胃癌发生、发展中起一定的作用,检测GKLF、IKLF的表达有助于判断胃癌患者的病情程度和预后。

Abstract:Objective To study the expression and clinical significance of gut-enriched Kruppel-like factor (GKLF) and intestinal-enriched Krappel-like factor (IKLF) in human gastric carcinoma cells and tissues. Methods Expressions of GKLF and IKLF in human normal gastric epithelium cell line (GES-1) and gastric carcinoma cell lines (MKN28,SGC7901,BGC823,HGC27),in 96 gastric carcinoma tissues and adjacent normal gastric tissues were detected by immunocytochemistry and immunohisto-chemistry. Results Expression of GKLF was seen in human normal gastric epithelium cell line. No expression of GKLF was seen in gastric carcinoma cell line. There was no expression of IKLF in human normal gastric epithelium cell line. There were expression of IKLF in gastric carcinoma cell lines. Positive expression rate of GKLF in the carcinoma tissues (20.8%) was significantly lower than that in adjacent normal gastric tissues(64.6%). Positive expression rate of IKLF in the carcinoma tissues(71.9%) was significantly higher than that in adjacent normal gastric tissues (21.9%). The expression of GKLF and IKLF was uncorrelated with age,gender,differentiation and Lauren's classification. They were correlated with lymph node involvement and clinical stage. Conclusion GKLF and IKLF play roles in the carcinogenesis and progression of gastric carcinoma. To examine the expression of GKLF and IKLF may be useful in judging the degree and prognosis of gastric carcinoma.
