槲皮素对人乳腺癌裸鼠移植瘤p21WAF1/CIP1的影响

摘    要:目的 研究槲皮素对人乳腺癌细胞株 MCF-7裸鼠移植瘤 p2 1W AF1/CIP1的影响。方法 采用 MCF-7细胞株移植成功的 2 4只裸鼠分为对照组、槲皮素组、阿霉素组及联合用药组 (槲皮素 +阿霉素 ) ,每组 6只。用原位杂交及免疫组化测定 p2 1W AF1/CIP1基因及蛋白的表达水平。结果  p2 1W AF1/CIP1m RNA表达水平槲皮素组及联合用药组高于对照组 (P<0 .0 5) ,p2 1W AF1/CIP1蛋白表达水平槲皮素组及联合用药组明显高于对照组 (P<0 .0 1)。结论 槲皮素可上调 MCF-7裸鼠移植瘤细胞的 p2 1W AF1/CIP1基因及蛋白的表达水平 ,减缓肿瘤生长

Abstract:Objective To study the effect of querce ti n on p21 WAF1/CIP1 in nude mice with transplanted breast carcinoma. Methods Twenty-four nude mice with transplanted tumor of MCF-7 were randomized into 4 groups with six mice in each group: control group, quercetin group, adriamycin group and combination group(quercetin+adriamycin). The gene an d protein expression of p21 WAF1/CIP1 were detected by in situ hybridizatio n and immunohistochemistry. Results For the expression of p21 WAF1/CIP1 mRNA, quercetin group and combination group were higher than contr ol group (P<0.05). For the expression of p21 WAF1/CIP1 protein, quercet in group and combination group were also higher than control group significantly (P<0.01). Conclusion Quercetin can up-regulate the gene a nd protein expression of p21 WAF1/CIP1 in nude mice with transplanted breas t carcinoma and inhibit the tumor growth.
