细胞外信号调节激酶ERK1/ERK2对胰腺癌生长的影响

摘    要:目的研究胰腺癌患者组织中及细胞系Pane-1及Jf-305中的ERK1/ERK2蛋白表达情况,以探讨其在胰腺癌发生、发展过程中的意义。方法应用免疫组织化学技术检测临床胰腺癌患者组织中ERK1/ERK2蛋白表达;Western blot方法观察胰腺癌细胞系Pane-1及Jf-305中的ERK1/ERK2蛋白表达。结果胰腺癌患者组织ERK1和ERK2蛋白均呈显著性高表达,阳性率分别为86.7%(39/45)和84.44%(38/45),而对照组织的阳性率分别为30%(6/20)和20%(4/20),胰腺癌组织和对照组织阳性率有显著差异;胰腺癌细胞系Panc-1及Jf-305中均有ERK1/ERK2蛋白的高表达。结论胰腺癌患者组织及细胞系中均有ERK1/ERK2蛋白强表达,ERK1和ERK2蛋白的过表达可能在人胰腺癌的发生、发展过程中起重要作用。

Abstract:Objective To observe the effect of ERK_1/ERK_2 on the growth of human pancreatic carcinoma tissues and cells.Methods The immunohistochemistry was used to detect the expression of ERK_1/ERK_2 in the pancreatic cancer tissues.The Western blot method was used to detect the expression of ERK_1/ERK_2 proteins in the pancreatic carcinoma cells.Results The immunohistochemistry showed that the positive rates of ERK_1/ERK_2 in the pancreatic cancer tissue were 86.7%(39/45)and 84.44%(38/45) respectively,whereas the positive rates of these proteins in the matched benignant pancreas disease were 30%(6/20)and 20%(4/20) respectively.The western blot showed that the expression of ERK_1 and ERK_2 proteins in the pancreatic carcinoma cells was also at a high level.Conclusion The expression of ERK_1/ERK_2 is significant at a high level in the pancreatic cancer tissue and also in the pancreatic carcinoma cells.The high expression of ERK_1/ERK_2 proteins may play an important role in the occurrence and development of pancreatic cancer.
