小檗碱对异烟肼致肝损伤模型小鼠的保护作用及其机制研究

摘    要:目的:观察小檗碱对异烟肼致肝损伤的保护作用及其机制。方法:NIH雄性小鼠30只,随机分为6组:正常对照组(生理盐水)、异烟肼组(180mg·kg-1)、联苯双酯组(异烟肼180mg·kg-1+联苯双酯150mg·kg-1)及小檗碱高、中、低剂量组(异烟肼180mg·kg-1+小檗碱100、50、25mg·kg-1),每日给药1次,连续5d。末次灌胃18h后处死小鼠,计算肝体指数,检测血清天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)活性及促炎因子白介素-6(IL-6)和抑炎因子IL-10含量,肝脏丙二醛(MDA)含量、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性。结果:与异烟肼组比较,小檗碱中、高剂量组肝体指数、AST、ALT、MDA、IL-6含量均显著降低(P<0.05或P<0.01),IL-10、SOD和GSH-Px水平显著升高(P<0.05或P<0.01)。结论:小檗碱可减轻异烟肼引起的肝损伤,其机制可能与抑制氧化应激和炎症反应有关。

Abstract:OBJECTIVE:To investigate the protective effects of berberine(Ber) against liver injury induced by isoniazid and its mechanism. METHODS:30 male mice were randomly divided into 6 groups:normal control group(normal saline) ,isoniazid group(180 mg·kg-1) ,bifendate group(isoniazid 180 mg·kg-1+bifendate 150 mg·kg-1) ,Ber high-dose,medium-dose and low-dose groups(isoniazid 180 mg·kg-1+Ber 100,50,25 mg·kg-1) . Mice were given medicine once a day for 5 days and sacrificed 18 h after the last intragastric administration. The hepatosomatic indices were calculated. The content of MDA,IL-6,IL-10 and the activities of ALT,AST,SOD and GSH-Px were detected. RESULTS:Compared with isoniazid group,the content of AST,ALT,MDA,IL-6 and hepatosomatic indices were decreased significantly(P0.05 or P0.01) ,and SOD,GSH-Px and IL-10 were increased significantly(P0.05 or P0.01) in Ber medium-dose and high-dose groups. CONCLUSION:Berberine has a protective effect on isoniazid-induced liver injury,the mechanisms may be related to inhibiting oxidative stress and inflammation.
