异莲心碱在大鼠体内的药动学研究

摘    要:目的:探讨异莲心碱在大鼠体内的药动学特性。方法:采用反相高效液相色谱法测定大鼠给予异莲心碱后血药浓度的变化。色谱柱为Hypersil C18(250 mm×4.6 mm,5μm),柱温为25℃,流动相为甲醇-乙腈-0.05 mol·L-1KH2PO4(22∶38∶40)(三乙胺调pH值至7.5),流速为1.0 mL·min-1,检测波长为280 nm。结果:线性回归方程为A=63.043c+3.937(r=0.999 8),异莲心碱检测浓度在0.50~40.00μg·mL-1范围内与其峰面积比值呈良好线性关系,最低检测浓度为0.10μg·mL-1,日内和日间精密度RSD均<4.10%,低、中、高浓度下样品的回收率均>91.20%。结论:本方法具有快速、简便、准确等优点,适用于异莲心碱在大鼠体内的药动学研究。

Abstract:OBJECTIVE:To establish a method for pharmaceutical study of isoliensine in rat.METHODS:RP-HPLC method was used to determine the concentration of isoliensine.The determination was performed on a Hypersil C18(250 mm×4.6 mm,5 μm) column with mobile phase consisted of methanol-acetonitrile-0.05 mol·L-1 KH2PO4(22:38:40)(pH adjusted to 7.5 with triethylamine) at a flow rate of 1.0 mL·min-1.The detection wavelength was set at 280 nm.RESULTS:The linear equation was A=63.043c+3.937(r=0.999 8).The linear range of isoliensine was 0.50~40.00 μg·mL-1.The lowest detection limit was 0.10 μg·mL-1.The RSD of intra-day and inter-day were below 4.10%.The recoveries of the control samples at low,medium and high concentrations were above 91.20%.CONCLUSION:The method is rapid,simple and accurate,and it is suitable for the pharmacokinetics study of isoliensine in rats.
