细菌性感染模型大鼠腹腔渗出液的上清液抗真菌作用研究

摘    要:目的:观察细菌性感染模型大鼠腹腔渗出液的上清液中抗菌物质对真菌的杀菌作用,为开发天然抗真菌药物提供依据。方法:取大鼠分为正常对照组和模型组,每组10只,模型组大鼠腹腔注射金黄色葡萄球菌(3.6×108cfu·mL-1)建立细菌性感染模型,建模后24h处死2组大鼠并收集腹腔渗出液,离心和处理后,观察渗出液、细胞液、上清液对白色念珠菌的杀菌作用,上清液对不同真菌的杀菌作用及温度对上清液杀菌作用的影响,并在显微镜下观察上清液对白色念珠菌的杀灭过程。结果:与正常对照组比较,模型组大鼠建模后24h的腹腔渗出大量渗出液,其中有大量白细胞,渗出液的上清液中含有大量蛋白(P均<0.01),具有与细胞液相似的抗真菌作用;上清液对白色念珠菌、热带念珠菌、光滑念珠菌具有强杀菌作用,可使白色念珠菌菌体萎缩进而破碎死亡;上清液经56℃水浴处理后杀菌作用几乎完全丧失。结论:细菌性感染模型大鼠腹腔渗出液的上清液中存在强抗真菌物质,具有强抗真菌作用,可能通过影响真菌的细胞壁/细胞膜而发挥作用,其中可能存在未被关注或未发现的抗菌蛋白/肽。

Abstract:OBJECTIVE: To investigate the fungicidal activity of the substance in peritoneal exudates supernatants in bacterial infection rats,and to provide reference for developing new natural fungicidal drugs.METHODS: Rats were divided into normal control group and model group with 10 rats in each group.Model group was injected with Staphyloccocus aureus intraperitoneally (3.6×108 cfu·mL-1) to establish bacterial infection model,and model rats were sacrificed 24 h after modeling to collect,separate and treat peritoneal exudates.The fungicidal activity of peritoneal exudation,cellular fluid,supernatant on Candida albicans was observed,and the fungicidal activity of supernatant on typical fungus and effect of temperature on fungicidal activity were observed.The duration of fungicidal activity was observed under microscope.RESULTS: Compared with normal control group,large amount of peritoneal exudates emerged in model group 24 h after modeling.There were a great of white cell in peritoneal exudates and protein (P0.01) in supernatants which possessed same fungicidal activity as cellular fluid.The peritoneal exudation supernatant possessed the most potent fungicidal activity on C.albicans,Oidium tropioale and Candida glabrata.C.albicans morphology changed gradually and then was expired due to disruption lastly after treatment with the supernatant,and the fungicidal activity of peritoneal exudates supernatants was destroyed dramatically after incubated at 56 ℃.CONCLUSION: The supernatant of peritoneal exudates from bacterial infection rats possesses potent fungicidal activity by influencing cell wall/cytomembrane of fungus.And there may be unconcerned and undiscovered antibacterial protein/peptide.
