布鲁氏菌强毒株16M和疫苗株M5差异探索

摘    要:目的探索布鲁氏菌强毒株16M和疫苗株M5之间的差异。方法用气相色谱测定两株菌的脂肪酸组成,用脉冲场凝胶电泳方法电泳两株菌的DNA酶切产物,寻找两者在脂肪酸组成和酶切电泳图谱上的差异。结果两株菌在脂肪酸的种类上一致,但含量有差异,强毒株16M脂肪酸16∶0含量较疫苗株M5高10%左右,而脂肪酸19∶0CYCLOω8c较疫苗株M5低15%左右。脉冲场凝胶电泳图谱显示在167.1kb和138.9kb之间强毒株16M比疫苗株M5多出一个条带。结论脂肪酸分析和脉冲场凝胶电泳技术都可以用于鉴别布鲁氏菌强毒株16M和疫苗株M5。

Difference of the vaccine strain M5 and the virulent strain 16M of Brucella species

Abstract:To explore the difference bctwccn the vaccine strain M5 and the virulent strain 16M of Brucella species, these two strains were inoculated on Bucella agar plates and incubated for 48 hours. After that, cells were harvested, saponificated, methylated and extracted to provide fatty acid mcthylester for gas chromatographic analysis. The DNAase Xba I cleavage products of these two strains were analyzed with pulsed field gel electrophoresis (PFGE). It was found that coincidence in the category of fatty acids existed in these two strains, but with difference in their contents. As demonstrated by fatty acids analysis, the content of fatty acid 18:0 w7c in the virulent strain 16M appeared to be higher than that of fatty acid 19 : 0 CYCLO w8c of the vaccine strain M5, in which that of the former was higher about 10% over the latter and the latter was lower about 15% lower than the former. As shown in the electrophoregram, there was a special band between 167.1 kb and 138.9 kb. It is apparent that both the analysis of content of fatty acid and PFGE analysis can be used to discriminate the vaccine strain and the the virulent strain of Brucella.
