疏肝健脾汤对肝纤维化大鼠基质金属2、TIMP-2影响的研究

目的：通过对肝纤维化大鼠基质金属蛋白酶及其抑制剂表达的影响,探讨疏肝健脾汤防治肝纤维化的机制。方法：雄性Wistar大鼠随机分为正常对照组、模型对照组、秋水仙碱预防组、秋水仙碱治疗组、疏肝健脾汤预防组、疏肝健脾汤治疗组,用猪血清诱导免疫损伤性肝纤维化模型,给予疏肝健脾汤进行防治,采用免疫组化方法,观测基质金属蛋白酶（MMP-2）及其抑制因子（TIMP-2）在各组大鼠肝脏组织中的表达。结果：模型组MMP-2较正常组有显著性差异（P〈0.05）,疏肝健脾汤各组MMP-2的表达较模型组有所降低（P〈0.05）,但高于正常组（P〈0.05）,低于秋水仙碱组（P〈0.05）;TIMP-2在模型组表达高于正常组（P〈0.05）,疏肝健脾汤各组表达低于模型组、秋水仙碱组,均有显著统计学意义（P〈0.05）。结论：疏肝健脾汤明显抑制MMP-2、TIMP-2的表达,提示疏肝健脾汤能够调节基质金属蛋白酶及其抑制因子的表达,促进细胞外基质的降解,从而实现对肝纤维化的防治作用。

The Effect of Shugan Jianpi Decoction on MMP-2 and TLMP-2 of Rats with Hepatic Fibrosis

Objective：To approach the mechanism of Shugan Jianpi Decoction on hepatic fibrosis by the effect of express about matrix metalloproteinase and catastaltica of rats with hepatic fibrosis.Methods：Male Wistar rats were randomly divided into normal control team,model control team,colchine prophylaxis team,colchine treatment team and Shugan Jianpi Decoction preventive team,Shugan Jianpi Decoction treatment team.The pig serum was used to induce immunologic injury hepatic fibrosis model and Shugan Jianpi San was used to prevent and cure.The immunohistochemistry was used to observe the express of MMP-2 and TLMP-2 of liver in every team.Results：Compared to nomal team,MMp-2 in model team had significant difference（P〈 0.05）,compared to model team,the express of MMP-2 in Shugan Jianpi Decoction team is lower（P〈 0.05）,but higher than nomal team（P〈 0.05）,lower than colchine team（P〈 0.05）,the express of TIMp-2 in model team is higher than nomal team（P〈 0.05）,alll had statistical significance（P〈 0.05）.Conclusion：Shugan Jianpi Decoction had the function to restrain the express of MMP-2 and TIMP-2 obviously.Its hint Shugan Jianpi Decoction can adjust the express of MPP and TIMP,advance degrade of ECM,to come true preventive and therapeutic effect on hepatic fibrosis.
