哮喘患者外周血淋巴细胞IL-5、IL-3、GM-CSFmRNA表达

目的 :探讨哮喘患者外周血单个核细胞(PBMC)IL 5、IL 3、GM CSFmRNA表达以及与慢性支气管炎患者的差异。方法 :选哮喘患者13例 (哮喘组 ) ,平均年龄(31.8±8.5)岁。慢性支气管炎患者9例 (慢支组 ) ,平均年龄(64.8±4.7)岁。健康献血员9例 (对照组 ) ,平均年龄(32.8±5.2)岁。抽取静脉血分离单个核细胞 ,用斑点印迹杂交法检测IL 5、GM CSF和IL 3mRNA的表达。结果 :哮喘组PBMCIL 5、GM CSF和IL 3mRNA表达的相对含量明显高于慢支组和对照组(P<0.01) ,慢支组与对照组相比无显著差异(P>0.05)。结论 :哮喘IL 5、GM CSF和IL 3mRNA表达明显增高 ,可能在哮喘发病中起重要作用。

Expression of IL-5, IL-3 and GM-CSF mRNA in peripheral blood mononuclear cells from patients with asthma

Objective: To determine the differences of IL 5, IL 3 and GM CSF mRNA expression in peripheral blood mononuclear cells (PBMC) between the patients with asthma and those with chronic bronchitis. Methods: The PBMCs were purified from the venous blood of 13 patients with asthma, 9 with chronic bronchitis and 9 healthy individuals. Then the expression of IL 5, IL 3 and GM CSF mRNA was determined with dot hybridization. Results: The level of IL 5, IL 3 and GM CSF mRNA expression was significantly higher in patients with asthma than in those with chronic bronchitis and healthy individuals (P<0.01). However, there was no marked difference between the patients with chronic bronchitis and healthy individuals. Conclusion: The pathogenic mechanism of asthma is different from that of chronic bronchitis. IL 5, IL 3 and GM CSF might contribute to the pathogenesis of asthma.
