电针对心理性应激致大鼠胃粘膜损伤的保护作用及与一些胃肠肽的关系

摘    要:目的:探讨电针对心理性应激状态下胃粘膜损伤的保护作用的机制。方法:将96只雄性SD大鼠随机分成3组,A:正常对照组;B:心理性应激组;C:心理性应激+针刺组。采用高压恒流脉冲刺激器制备心理性应激模型。电针刺激大鼠双侧足三里,用Chen法测定胃粘膜损伤指数并计算保护率。用放免分析法、生化法同步测定各组血浆中SS,NOS,CGRP,VIP的含量。结果:心理应激条件下,B组胃粘膜出现损害、甚至产生溃疡,且随着应激反应程度的不断增强,胃粘膜损害的程度不断加大,而C组胃粘膜损伤明显减轻,且随着时间的延长,这种效果更加明显。胃粘膜保护率也增加。大鼠血浆SS的浓度B组显高于A组及C组(P<0.01),B组与C组SS有明显差异(P<0.01)。NOS的活性,B组明显高于A组(P<0.01),C组与B组也有明显差异(P<0.05),但是,A组与C组二无差异(P>0.05),CGRP的水平在B组发生很大的变化。随着应激程度的不断加强,CGRP的水平不断升高,与A组相比有明显差异(P<0.01)。然而,C组与A组相比无明显差异(P<0.05),但与B组相比差异显(P<0.01)。心理性应激状态下,VIP水平的明显升高。B组与A组相比差异明显(P<0.01)。C组VIP的水平与A组无明显差异(P>0.05),而与B组差异明显(P<0.05)。结论:心理性应激导致胃粘膜的操作及电针穴位对其产生调节防治作用的机制与某些胃肠道激素或神经肽密切相关。

Abstract:AIM To test whether the acute gastric mucalsal lesion can be caused by psychological stress and to explore protective effects of electroacupuncture on rats under psychological stress. METHODS Animal model was constructed with impulsive stimulator of high voltage and stable currency stimulating the Chusanli with electroacupuncture in rats. Ninety six male sprague Dauley rats were randomly divided into three groups: (A) control group, (B) psychological stress group and (C) electroacupuncture group. The gastric mucosal injury index was calculated with CHUN method. By using radioactive immunon assay and biochemical methods, the change in contents of plasma SS, NOS, CGRP, VIP was simultaneously measured in different lab conditions in lab rats. RESULTS Under psychological stress, gastric mucosal lesion appeared, even did ulcer. The degree of gastric mucosal lesion became more and more serious, and lesion index increased gradually with the continuous psychological stress. But gastric mucosal lesion alleviated significantly under psychological stress with electroacupuncture stimulation. And the effect was more significant with the continuous electroacupuncture stimulation. There was significant difference in lesion indexes between EA group and psychological stress group ( P <0.01). The protective rate increased gradually with the electroacupuncture stimulation. We measured the plasma SS contents which were significantly different in control group and psychological stress group ( P <0.01), and psychological stress group and EA group were the same as that ( P <0.01). The activity of NOS in plasma was very high under psychological stress. There was difference between those of them ( P <0.01). The activity of NOS in plasma of EA group was similar to the control group ( P >0.05). But it was significantly different with psychological stress group ( P <0.01). The activity of CGRP in plasma of rats was gradually developed by psychological stress with degree of psychological stress. There was difference between those of them ( P <0.01). The activity of CGRP in plasma of rats under psychological stress was much higher than normal rats. But the activity of CGRP in plasma of EA group was similar to the control group ( P >0.05). The activity of VIP in plasma of lab rats was approximately similar to the CGRP. CONCLUSION Psychological stress can lead to gastric mucosal lesions, and the correlation is severe. Stimulating Chusanli can impose protective and therapeutic effects on them. This effect is probably related to the changes in plasma gut hormones.
