牛磺酸对肝癌大鼠肝细胞保护机制的研究

摘    要:目的探讨牛磺酸(taurine,Tau)对肝癌大鼠肝脏细胞的保护作用。方法将24只Wistar肝癌大鼠随机分成对照组和牛磺酸低、中、高剂量组(饮水中分别添加0.5%、1%、1.5%牛磺酸),连续喂养5w后取血测定血清丙氨酸氨基转移梅(ALT)、天门冬氨酸氨基转移酶(AST)含量,并测定肝脏超氧化物歧化酶(SOD)活性、谷胱甘肽过氧化物酶(GSH-Px)活力及丙二醛(MDA)含量。结果血清ALT、AST含量明显升高,与对照组比较,有显著性差异(P〈0.05,P〈0.01),肝脏SOD活性、GSH-Px活力与正常对照组比较,有显著性差异(P〈0.05,P〈0.01),随着牛磺酸摄入量的增加,各组MDA含量呈逐渐下降趋势,与正常对照组比较,有显著性差异(P〈0.05,P〈0.01)。结论牛磺酸可通过抗氧化损伤作用对肝癌大鼠肝细胞起保护作用。

Abstract:Objective To observe the protection of taurine(Tau)on hepatic cells in rats with hepatic carcinoma.Methods 24 Wister rats with hepatic carcinoma were divided randomly into control group,and groups of Tau in low dose,moderate dose and high dose(0.5%,1%,1.5% of Tau was added in drinking water successively).After five weeks of feeding,the content of alanine amiotransterase(ALT)and aspartate transaminase(AST)in serum and the activity of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and maleic dialdehyde(MDA)in the liver were detected.Results Compared with control group,significant differences were presented in the contents of ALT and AST in serum(P〈0.05,P〈0.01).The significant differences were obtained in SOD and GSH-Px activity in the liver(P〈0.05,P〈0.01)compared with those in normal control group.With the increase of intake volume of Tau,MDA content in every group was decreased in tendency and significant differences had been obtained compared with normal control group(P〈0.05,P〈0.01).Conclusion Taurine protects hepatic cells in rats with hepatic carcinoma through its resistance to oxidative damage.
