纤维蛋白贴安全性药理实验研究

摘    要:目的研究纤维蛋白贴的安全性药理,为新药研发提供依据。方法通过小鼠自发活动实验、协调力实验和戊巴比妥钠催眠实验来考察纤维蛋白贴对中枢神经系统的影响;通过麻醉Bealge犬实验考察纤维蛋白贴对呼吸系统和心血管系统的影响;通过正常大鼠和肝部份切除大鼠分别考察纤维蛋白贴对泌尿系统和凝血功能的影响。结果纤维蛋白贴剂量为1/10贴每只小鼠时对小鼠自主活动次数、机体协调能力、戊巴比妥钠诱导的小鼠睡眠时间均未见明显影响;在4贴每犬时对麻醉Beagle犬呼吸及心血管系统无毒理学意义的改变;在1/2贴每只大鼠剂量下,对大鼠凝血功能和泌尿系统亦无毒理学意义的改变。结论在本研究条件下,纤维蛋白贴对中枢神经系统、呼吸系统、心血管系统、泌尿系统和凝血功能均未见明显影响,安全性较好,推测其具有良好的市场应用前景。

Safety Pharmacology Research of Fibrin Patch

Abstract:Objective To study the safety pharmacology of fibrin patch, and provide foundation for clinical research. Methods The effect on central nervous system was observed by autonomic activities experiment, mice' concordant ability experiment and hypnotize experiment were induced by pentobarbital. The effects on respiratory system and cardiovascular system were observed in anesthetized Beagle dogs. The effect on urinary system and blood clotting were observed in normal rats. Results Fibrin patch at the dose of 1/10 patch of each mouse showed no influence on the mice' autonomic activity, concordant ability and Somnus time induced by pentobarbital. Fibrin patch at the close of 4 patch of each dog showed no influence on the central nervous system, respiratory system, circulatory system. Fibrin patch at the dose of 1/2 patch of each rat showed no influence on the urinary system and the function of blood clotting. Conclusion In this study, Fibrin patch has no toxicity on the nervous system, respiratory system, cardiovascular system, urinary system and the function of blood clotting. The safety of fibrin patch can be believed fine, and it may have a good market prospect.
