高压氧对脑缺血再灌注海马CA1区神经元凋亡的作用

摘    要:目的研讨高压氧(HBO)对脑缺血再灌注损伤的疗效及其机制,为临床HBO治疗疾病提供理论依据。方法实验动物用沙土鼠20只,雌雄不限。采用随机数字法将实验动物分为正常对照组、缺血组、0.15MPaHBO治疗组、0.25MPaHBO治疗组、0.25MPa压力空气(hyper-baricair,HBA)对照组5组(n=4)。应用TUNEL检测技术,对沙土鼠前脑缺血20min后再灌注3d模型,用高压氧(hyperbaricoxygen,HBO)治疗连续3d,观察HBO作用下海马CA1区神经元凋亡变化。结果沙土鼠脑缺血再灌注3d后海马CA1区大量神经元凋亡犤(163±15)/mm2犦,HBO治疗组凋亡细胞数犤0.15MPaHBO治疗组为(99±12)/mm2,0.25MPaHBO治疗组为(73±11)/mm2犦明显减少(P<0.01),0.25MPa空气对照组凋亡细胞数为(151±13)/mm2,以0.25MPaHBO治疗组为佳。结论HBO治疗对海马神经元损伤有保护作用,减少神经元凋亡,为高压氧治疗缺血性损伤的疗效机制之一。

Abstract:Aim To study the mechanism of hyperbaric oxygen ( HBO ) on the cerebral ischemia and reperfusion injury and to provide theoretical basis for hyperbaric oxygen treatment ( HBOT ).Methods 20 gerbils weighed 60-90 g were divided by random number table into normal control,ischemia,0.15 Mpa HBO treatment,0.25 Mpa HBO treatment,0.25 Mpa hyperbaric air(HBA) control groups(n=4 in each group).The effect of 0.15 Mpa and 0.25 Mpa HBO treatment ( 60 min/d, for 3 successive days) on ischemia reperfusion-induced apoptosis in gerbil hippocampus CA1 were examined by using TUNEL staining method.Results Large amount of apoptotic neurons were found in CA1 area of hippocampus of gerbils after 3 days of ischemia and reperfusion (163± 15)/mm2].The number of apoptotic neurons in 0.25 Mpa HBO treatment group (73± 11)/mm2] was obviously decreased than that in 0.15 Mpa HBO treatment group (99± 12)/mm2, P< 0.01].The number of apoptotic neurons was (151± 13)/mm2.Conclusion HBOT is effective in reducing neuronal apoptosis and in protection on neurons from injury of ischemia reperfusion, and 0.25 Mpa is an appropriate pressure for HBOT.
