| 摘 要： | 目的：探讨人类端粒酶RNA的反义寡核苷酸对K562细胞增殖的影响。方法：采用MTT和试验和平板集落形成实验观察反义寡核苷酸作用后K562细胞的生长情况，采用流式细胞仪分析细胞周期的改变，采用电镜观察细胞超微结构的改变，用PCR－ELISA分析细胞端粒酶活性的改变。结果：端粒酶RNA的反义寡核苷酸作用K562细胞后，细胞超微结构和细胞周期发生明显改变。端粒酶活生到明显抑制，细胞增殖受到明显抑制。结论：靶向于端粒酶RNA的反义寡核苷酸抑制白血病细胞K562的增殖。

| Abstract: | Objective:To investigate the ef fects of AS-ODN targeting against telomerase RNA on the proliferation in K56 2cell line.Methods:The growth of K562cells were exam-ined by MTT test and formed colonestest;cell cycle distribution were detected by flow cytometry ana lysis;morphologic changes was examined by electronmicroscope and telomerase act ivity were analysed by PCR-ELISA.Results:Morphologic changes and cell cycle d istribution were signifi-cantly changed and telomerase activity was markedly in hibited in K562cells treated with AS-ODN targeting against telomerase RNA,a nd cell growth was significantly inhibited.Conclusion:AS-ODN targetingagainst telomerase RNA can inhibit the proliferation in K562cell line. |
