胰岛素对血管平滑肌细胞增殖及原癌基因c-fos,c-myc异常表达的实验研究

摘    要:我们观察了不同浓度的胰岛素对体外培养人血管平滑肌细胞增殖的影响,及其该过程中原癌基因c-fos,c-myc的表达。结果表明,胰岛素能促进血管平滑肌细胞(SMC)增殖,即:SMC数目的增多,SMCDNA的合成;胰岛素能促进原癌基因c-fos和c-myc的异常表达;且以上作用均存在着剂量-效应关系。这些结果提示,高胰岛素血症促进动脉粥样硬化(AS)发生、发展可能是通过促进某些原癌基因异常表达,进而促进SMC增殖而实现的。这也许是某些基因治疗AS的基础。

Abstract:To test whether or not insulin can stimuate c fos,c myc expression and proliferation of vascular smooth muscle cell (VSMC),we studied the growth effects of insulin as measured by stimulation of c fos,c myc, DNA synthesis and cellular proliferation in the presence different concentrations of insulin.These findings demonstrated that insulin is capable of increasing c fos,c myc mRNA,DNA synthesis and cell number in cultured human artery SMC,and these changes are dose effect relationship .So we postulate that insulin stimulates c fos,c myc expression and proliferation of VSMC is one way of hyperinsulinemia contributing to cardiovascular disease.
