脑泰方对局灶性脑缺血大鼠血清MMP-9、PA及TIMP-1表达的影响

摘    要:目的研究脑泰方治疗对局灶性脑缺血大鼠血清中基质金属蛋白酶9(MMP-9),MMP-9前体纤溶酶原激活物(plasminogen activator,PA)及内源性抑制剂1(TIMP1)的作用。方法随机将SD大鼠分为假手术组、空白对照组、脑泰方低、中、高剂量组(3,9,27g/kg),marimastat(BB-2516)组(6.25mg/kg)ig。各组大鼠预处理ig给药连续3d,每日1次,后采用大脑中动脉栓塞法(Middle cerebral artery occlusion,MCAO)行大鼠局灶性脑缺血模型制备,术后连续ig给药3d。采用ZeaLonga神经学评分标准记录动物的行为活动,采用ELISA法检测血清中MMP-9、PA及TIMP1的表达。结果脑泰方高剂量组神经学评分较空白对照组有显著降低(P〈0.05);脑泰方高、中、低剂量组血清中MMP-9、PA的表达显著降低(P〈0.05),脑泰方高、中、低剂量组TIMP1表达明显升高(P〈0.05)。结论脑泰方对局灶性脑缺血导致的血脑屏障损伤有一定保护作用。

Effects of Naotaifang on Serum MMP-9,PA and TIMP-1 after Focal Cerebral Ischemia in Rats

Abstract:Objective To observe Effects of NaoTai Fang on serum MMP-9, PA and TIMP-1 after Focal Cerebral Ischemia in Rats. Methods The rats were randomly allotted 5 groups as following .. sham operation group, model group, low dose group of NTF (3g/kg), medial dose group of NTF (gg/kg), large dose group of NTF (27g/kg), marirnastat (BB-2516) group (6.25mg/kg) . After 3 days of corresponding ther- apy by intragastric administration once every day, The regional cerebral ischemia model was reproduced by Middle cerebral artery occlusion (MCAO) with suture method. Following 3 days, the rats treated with previous method. The third day, neuroethology expression was detected by Zea Longa neurological score. The contents of MMP-9, PA and TIMP1 proteins in serum were measured by enzyme-linked immuno sorbent assay (ELISA) method with marketable kit. Results In large dose group of NTF the expression of neurological score was significantly decrease than model group ( P 〈0. 05) ; In low , medial , large dose group of NTF and marimastat (BB-2516) group the expressions Of MMP-9. PA were less significantly decreased than model group ( P 〈0.05), and expressions Of TIMP1 were significantly increased than model group ( P 〉 0. 05) . Conclusion The mechanism of NTF protecting brood brain barrier (BBB) against injure of cerebral ischemia may be involved in amelio-rating MMP, PA and increasing TIMP1 expression.
