原发性肾病综合征模型小鼠IL-18水平表达的研究

摘    要:目的 探讨白细胞介素-18(interleukin-18,IL-18)在微小病变型肾病综合征(minimalchangenephropathy,MCN)模型小鼠免疫失调过程中的作用。方法采用阿霉素单次尾静脉注射昆明种小鼠,诱导形成MCN模型,设置生理盐水对照组进行比较。测定两组小鼠第1、2、4、6周各时期24小时尿蛋白量(24hoursurinaryproteinquantity,24HUPQ)水平;于第42天检测两组小鼠血清总蛋白(totalprotein,TP)、白蛋白(albumin,ALB)、尿素氮(bloodureanitrogen,BUN)、肌酐(creatinine,Cr)、甘油三酯(triglyceride,TG)、总胆固醇(totalcholesterol,TC)的含量;并采用ELISA法检测两组小鼠血清、肾组织匀浆上清、脾淋巴细胞培养液上清IL-18、INF-y,TNF—a及IL-4水平。结果模型组小鼠第2、4、6周24HUPQ值均显著高于对照组,且差异均有统计学意义(P均〈0.01)。模型组的TP、ALB水平较对照组均显著降低,TC、TG水平均显著升高,且差异均有统计学意义(P均〈0.01),而BUN、Cr水平两组间差异均无统计学意义(P均〉0.05)。模型组小鼠血清、肾组织匀浆上清、脾淋巴细胞培养液上清IL—18水平、INF-y和TNF-a的水平均显著高于对照组,而IL-4的水平则低于对照组,且差异均有统计学意义(P均〈0.01)。结论IL-18是参与小鼠MCN发病过程中重要的免疫因子。

Abstract:Objective To discuss the effect of interleukin-18(IL-18) in immune function disturbance of mice model with minimal change nephropathy (MCN). Methods MCN models were induced in Kunming type mice by injected adriamycin through tail vein, at the same time, saline solution was given to other mice in control group. The levels of 24 hours urinary protein quantity (24 HUPQ) in the 1st, 2nd, 4th, 6th weeks were detected. At around 42nd day, the levels of total protein(TP), albumin(ALB), blood urea nitrogen(BUN), creatinine (Cr), triglyceride(TG), total cholesterol(TC) were detected. At the same time, the levels of IL-18, INF-7, TNF-a and IL-4 of the serum, kidney tissue homogenate supematant and splenic lymphocyte culture fluid supematant were detected by ELISA method. Results The levels of 24 HUPQ in 2, 4, 6 weeks in model group were higher than in control group and the differences all had statistical significance (Pall〈 0.01). The results of serum TP, ALB in model group were lower than in control group, while the levels of TC, TG were higher than in control group and the differences all had statistical significance (Pall〈 0.01). There were no statistical significance in the differences of the levels of Cr and BUN between the two groups (Pall〉 0.05). The levels of IL-18, INF-y, TNF-a in serum, kidney tissue homogenate superuatant and splenic lymphocyte culture fluid supernatant in model group were all higher than in control group, while the IL-4 levels were opposite and the differences all had statistical significance(Pall〈 0.01). Conclusion IL-18 is the important immune factor in the development of MCN.
