大鼠血管平滑肌细胞分离培养的探讨

目的：培养大鼠血管平滑肌细胞。方法：手术显微镜下分离主动脉和肺动脉，再用翻转干涸法进行操作。结果：显微镜下分离组织，岢基本除净动脉纤维脂肪层和外膜，再用刀片轻刮内膜，可保证所贴组织块为地动脉中膜。传至第三代时，用台盼蓝检查细胞传代成活率９５％，光镜、电镜和免疫组化鉴定培养细胞，培养细胞纯度９６％。结论：贴块片简便易行、经济、又可以防止膜损伤，可为血管病理变化研究提供适宜的细胞。

Culture of rat pulmonary artery smooth muscle cells

Objective: To culture rat vascular smooth muscle cell (VSMC). Methods: Pulmonary artery were separated carefully and cut into fine tissue pieces. These pieces used for culture of VSMC by method of adhesion to wall. Results: Cell cultured were typical form of VSMC under light microscope, electron microscope and immunohistochemical test. The cell viability is 95 % in subculture by trypan blue. The cell purity is 96% by immuohistochemical method. Conclusion: This method is simple and convenient. It provides ideal materials for vascular physiopathologic studies.
