红车轴草总黄酮体外抗氧化作用的研究

摘    要: 目的 探讨红车轴草总黄酮体外抗氧化作用。 方法 采用分光光度法测定 H2O2 诱导红细胞溶血作用,用 2- 硫代巴比妥酸法测定小鼠红细胞膜脂质及肝组织匀浆脂质过氧化产物丙二醛( MDA )含量。 结果 红车轴草总黄酮在 0.2~1.6 mg·L-1 内能抑制 H2O2 诱导红细胞溶血和红细胞膜脂质过氧化作用, IC50 分别为 0.93 及 0.92 mg·L-1 ,均呈较好的量效关系;红车轴草总黄酮在 4~32 mg·L-1 内能抑制肝组织脂质过氧化产物的形成, IC50 为 17.5 mg·L-1 。 结论 在一定浓度范围内红车轴草总黄酮体外具有明显的抗氧化作用。

Abstract:OBJECTIVE To study the anti-oxidation of the flavonoids in the red clover in vitro. METHODS Spectrophotometry was used to measure the hemolysis of the erythrocytic induced by H2O2 and TBA colorimetry to study the content of lipid peroxidation products (MDA) in the erythrocytes and the liver homogenates of mice. RESULTS The results indicated that the flavonoids of the red clover at the concentration of 0.2-1.6 mg·L-1 significantly inhibited the erythrocytic hemolysis induced by hydrogen peroxide and the lipid peroxidation of erythrocytic membranes.50% inhibitory concentrations (IC50) were 0.93 and 0.92 mg·L-1 respectively, showing a appropriate concentration-effect relationship. And they obviously prevented the formation of MDA lipid peroxidation in homogenates of mouse liver at the concentration of 4-32 mg·L-1, with IC50 of 17.5 mg·L-1. CONCLUSION The flavonoids of the red clover have a significant antioxidation effect within a range of certain concentration in vitro.
