山茱萸炮制前后的HPLC指纹图谱研究

摘    要: 目的建立山茱萸炮制前后的高效液相(HPLC)指纹图谱。方法选择山茱萸生、制品各12批,以体积分数80%甲醇溶液超声提取制备溶液,应用Agilent Zorbax C18(4.6mm×250mm,5μm)柱,二极管阵列检测器(DAD),以0.1%磷酸溶液和乙腈为流动相,梯度洗脱,流速1mL·min-1,柱温30℃,检测波长240nm。结果山茱萸生品样品中共检测到19个共有色谱峰,可以指认5个共有峰;山茱萸制品样品中共检测到23个共有色谱峰,可以指认6个共有峰。结论从整体上显示生、制品的特征,为山茱萸生、制品的质量控制提供有效手段。

Abstract:OBJECTIVE To establish the fingerprint of crude and processed Cornus officinalis by HPLC. METHODS The 12 batches of crude drug and 12 batches of processed products were extracted in 80% methanol ultrasonic extraction, respectively. Subsequently, the samples solution was separated on an Agilent Zorbax C18 column (4.6 mm×250 mm, 5 μm) and detected by DAD.Mobile phase was composed of aqueous phosphoric acid (0.1%) and acetonitrile using a gradient elution. Analysis was performed at 30 ℃ with a flow rate of 1 mL·min-1 and UV detection at 240 nm. RESULTS There were 19 common peaks in crude drug and 5 peaks identified. There were 23 common peaks in processed products and 6 peaks identified. CONCLUSION The fingerprint of crude and processed Cornus officinalis could be applied to the intrinsic quality control of crude and processed Cornus Officinalis.
