雷公藤药材HPLC指纹图谱的研究

摘    要: 目的建立雷公藤药材的色谱指纹图谱。方法采用高效液相色谱法分析,色谱柱:Kromasil C18(4.6mm×250mm,5μm),流动相:乙腈-水(0.1%磷酸)进行梯度洗脱,柱温35℃,检测波长267nm,分析时间80min;流速1.0mL·min-1,测定10批药材的色谱图,确立标准指纹图谱。结果指纹图谱标出雷公藤药材的29个共有峰,以化学对照品结合HPLC-MS鉴定了其中18个峰。结论该分析方法的稳定性、精密度、重现性较好,为雷公藤药材的质量控制提供了依据。

Abstract:OBJECTIVE To develop a HPLC fingerprint method for the quality control of Tripterygium wilfordii.METHODS A HPLC analysis was carried out on a Kromasil C18 column(4.6 mm×250 mm,5 μm)with the mobile phase consisting of acetonitrile(A)-water (0.1% phosphoric acid) in gradient mode.The detection wavelength was set at 267 nm and the column temperature was kept at 35 ℃.The flow rate was 1.0 mL·min-1 and the analytic time was 80 min.The ten batches of samples were determined,for establishing the standard HPLC fingerprint of T. wilfordii.RESULTS 29 Peaks were identified as common peaks,18 peaks of them were identified as active compounds by the reference compounds and HPLC-MS.CONCLUSION The method was in good reproducibility,precision and stability.The present research provides new evidence for the quality control of T. wilfordii.
