肉桂酸的代谢与CYP450酶的相关性研究

摘    要: 目的通过离体肝匀浆实验法来研究肉桂酸的代谢转化过程,探讨参与肉桂酸代谢的CYP450亚酶,为临床上合理用药提供科学依据。方法应用高效液相色谱法检测大鼠肝匀浆温孵液中剩余肉桂酸的含量,分析各诱导和抑制剂对肉桂酸代谢的影响。结果特异性CYP1A诱导剂β-NF组中肉桂酸代谢速率明显高于对照组,而主要诱导CYP2B的PB组和CYP3A的DEX组与对照组比较无明显区别;特异的CYP1A抑制剂α-NF则显著抑制肉桂酸的代谢。结论CYP1A是介导肉桂酸代谢转化的主要酶系,肉桂酸与抑制或诱导CYP1A酶的药物合用可能存在相互作用。

Abstract:OBJECTIVE To study the metabolism of cinnamic acid in homogenized liver of rats, and to evaluate the effect of the CYP450 enzyme, which participate in metabolism of cinnamic acid,and then to supply the evidence for rational administration. METHODS The content of cinnamic acid in homogenized liver was measured by the reversed phase HPLC to analyze the effects of inductors and inhibitors on metabolism of cinnamic acid. RESULTS The metabolic rate of cinnamic acid in β-NF group, the specific CYP1A inductor β-NF group, was significantly higher than that in control group. CYP2B inductor PB group and CYP3A inductor DEX group, had no significant difference compared with control groups. Further more,α-NF, the CYP1A inhibitor, inhibited the metabolism of cinnamic acid significantly. CONCLUSION CYP1A is the major enzyme in metabolism of cinnamic acid. The medicine, which have inducible and inhibitory effects on CYP1A, may have interaction with cinnamic acid.
