摘    要: 目的分别考察橙皮苷在大鼠各肠段的吸收动力学特征及不同药物浓度、不同pH值灌流液对其吸收的影响。方法采用大鼠在体肠单向灌流实验方法,用HPLC对灌流液中的橙皮苷进行分析。结果橙皮苷在肠道中的吸收按回肠、空肠、十二指肠、结肠的顺序依次下降,其中在结肠中的吸收显著低于其他肠段(P<0.05);pH5.0、6.8的酸性条件下肠道对橙皮苷的吸收显著高于pH7.8弱碱条件(P<0.05);随着橙皮苷浓度的增加其KaPapp均逐渐下降,其中质量浓度为2mg·L-1的溶液Papp显著高于其他2个浓度(P<0.05)。结论橙皮苷在全肠段均有吸收,但在回肠中吸收较好;弱酸性环境易于橙皮苷吸收;橙皮苷在大鼠肠道的吸收存在饱和现象,其在机体内的转运可能存在主动转运或促进扩散。

Abstract:OBJECTIVE To investigate the absorption kinetics of hesperidin at different intestinal segments of rats and the influence of pH and the drug solution concentration on the absorption kinetic.METHODS In situ single pass perfusion model and HPLC method were used to investigate the absorption and determine the concentration of hesperidin in solution.RESULTS The absorption of hesperidin decreased followed the sequence of ileum,jejunum,duodenum and colon.The absorption in colon was obviously lower than those in other intestinal segments(P<0.05).The absorption of hesperidin under acid condition at pH 5.0 and 6.8 was obviously higher than weak alkaline condition at pH 7.8(P<0.05).With the increase of hesperidin concentration,Ka and Papp decreased,and Papp of 2 mg·L-1 solution was obviously higher than those of the other two concentrantion.CONCLUSION Hesperidin can be absorbed in whole intestinal segments,but best in ileum.The absorption can be better under acid condition.The transport mechanism of hesperidin in vivo is similar to active transport or facilitated diffusion.
