人白血病细胞系(J6-1)膜相关调节因子(MAF-J6-1)的分离与性质

我们由人白血病细胞系J6-1发现了对自身有增殖刺激作用的膜相关生长调节因子(MAF-J6-1)。经Sephadex G-100凝胶过滤及连续两次快速蛋白液相层析(FPLC,Mono Q)将其分离。理化性质研究的结果表明,它是一种对酸、碱和热相对稳定的蛋白,分子量约50kD。抗M-CSF单抗与MAF-J6-1的中和试验结果表明,MAF-J6-1属M-CSF类。

ISOLATION AND CHARACTERIZATION OF MEMBRANE ASSOCIATED REGULATING FACTOR FROM HUMAN LEUKEMIC CELL LINE J6-1

J6-1 leukemic cell line was established in our laboratory from AMML patient and its proliferation depends on the cell-cell contact. The J6-1 cell membrane lysate, but not conditioned medium, showed activity for J6-1 cell's proliferation. Recently, the J6-1 membrane associated factor, stimulating proliferation of J6-1 cells, was discovered and designed as MAF-J6-1. After fractionation via Sephadex G-100 and sequential two steps FPLC Mono Q, it was purified over 333-fold. MAF-J6-1 was moderately stable for heat, acid and alkali, the molecular weight was 50 kD as estimated by SDS-PAGE. The neutralization test was performed with the McAb of M-CSF. The result showed that the MAF-J6-1 might be a membrane bound M-CSF.It is suggested that J6-1 leukemic cells might exist contact stimulating autocrine mechanism for proliferation, and the membrane bound M-CSF plays an important role in the contact stimulation.
