摘 要: | 利用乳糖诱导工程菌pET(32a+)-PTH(1-34)/BL21(DE3) 高效表达人甲状旁腺激素1-34融合蛋白.并对乳糖浓度、诱导温度、诱导时机等参数进行优化,得到目的蛋白高表达乳糖诱导高密度发酵工艺.三批实验结果表明,人甲状旁腺激素1-34融合蛋白表达量在33%左右,工程菌生物量在55 g/L左右.

Abstract: | An engineering bacterial strain pET(32a+)-PTH(1-34)/BL21(DE3) which can express the fusion protein of human parathyroid hormone(1-34) has been studied for high-density fermentation induced by lactose in this paper.Through the optimization of lactose concentration,the temperature of induction and the time of induction,we got a high-density fermentation process which can high performance express target protein induced by lactose.Three lots of engineering bacterial strain were cultured,and the results showed that the expression of human parathyroid hormone 1-34 fusion protein was 33% and the weight of bacteria strain was 55g/L approximately. |
