文拉法辛手性拆分影响因素的分析

摘    要:目的研究文拉法辛手性拆分的影响因素。方法采用万古霉素手性柱对文拉法辛进行手性拆分,流动相为甲醇-醋酸胺缓冲液,用甲酸调缓冲液pH值,考察其对手性拆分参数的影响。利用串联质谱ESI电离正离子模式、多反应监测(MRM)扫描方式监测。结果当流动相比例为92:8,缓冲液pH值为3.33时,文拉法辛拆分结果最佳。结论缓冲液pH值对文拉法辛手性拆分的影响较大,是所有参数中应该最先优化的参数。

Abstract:Objective To determine the impact of mobile phase on the chiral separation of venlafaxine by an HPLC-MSMS method. Methods The venlafaxine enantiomers were separated by the vancomycin chiral volumn. The mobile phase consisted of methanol and ammonium acetate buffer whose pH was modified by the formic acid. The compounds were detected by the mass spectrometry under the conditions of ESI+ source and multiply reaction monitor model (MRM). Results When the mobile phase consisted of 92% methanol and 8% ammonium acetate buffer (pH=3.33), the venlafaxine enantiomers were separated completely. Conclusion The pH value of the mobile phase has the greatest effect on the chiral separation of venalfaxine.
