C反应蛋白基因单核苷酸多态性与脓毒症易感性及其严重程度的关系

摘    要:【】目的:研究C反应蛋白(CRP)基因rs1205位点(+1846 C>T)和rs3093062位点(?409G > A)单核苷酸多态性(single nucleotide polymorphism, SNP)与脓毒症易感性及其严重程度的关系。方法:采用直接测序法检测125例脓毒症患者和125例健康对照组CRP基因rs1205位点和rs3093062位点基因型,采用酶联免疫吸附法(ELISA)检测受试者血清CRP蛋白水平。结果:CRP基因rs1205位点TT基因型是脓毒症的危险因素(OR=3.02,95%CI=1.18-7.87,P=0.01)。CRP基因rs3093062位点GA和AA基因型是脓毒症的危险因素(OR=2.68,95%CI=1.19-4.83,P<0.01;OR=4.18,95%CI=1.59-11.29,P<0.01)。不同性别、年龄受试者CRP基因rs1205位点和rs3093062位点基因型频率之间的差异没有统计学意义(P>0.05)。CRP基因rs1205位点TT基因型以及rs3093062位点AA基因型患者脓毒症较为严重(P>0.05)。CRP基因rs1205位点和rs3093062位点突变型受试者血清CRP蛋白水平较高,差异有统计学意义(P<0.05)。结论:CRP基因rs1205位点+1846 C>T突变和rs3093062位点?409G > A突变与脓毒症易感性及其严重程度相关。

Relationship between single nucleotide polymorphism of C-reactive protein gene and susceptibility and severity of sepsis
Abstract:Objective: To study the relationship between the single nucleotide polymorphisms of the C-reactive protein (CRP) gene rs1205 (+1846 C>T) and rs3093062 (?409G > A) and the susceptibility and severity of sepsis. Methods: The genotypes of CRP gene rs1205 and rs3093062 were detected by Sanger sequencing method in 125 sepsis patients and 125 healthy controls. Serum CRP protein levels were detected by enzyme-linked immunosorbent assay (ELISA). Results: The TT genotype of the rs1205 locus of the CRP gene was a risk factor for sepsis (OR=3.02, 95% CI=1.18-7.87, P=0.01). CRP gene rs3093062 locus GA and AA genotype were risk factors for sepsis (OR=2.68, 95% CI=1.19-4.83, P<0.01; OR=4.18, 95%CI=1.59-11.29, P<0.01) . There was no significant difference in genotype frequencies between CRP gene rs1205 locus and rs3093062 locus in different sexes and ages group (P>0.05). Patients with CRP gene rs1205 TT genotype and rs3093062 AA genotype had severe sepsis (P>0.05). Serum CRP protein levels were higher in subjects with CRP gene rs1205 and rs3093062 mutations, and the differences were statistically significant (P<0.05). Conclusion: CRP gene rs1205 locus +1846 C>T mutation and rs3093062 locus ?409G > A mutation are associated with susceptibility and severity of sepsis.
