佛山汉族高血压患者亚甲基四氢叶酸还原酶基因的分布

目的 探讨广东佛山汉族地区高血压患者亚甲基四氢叶酸还原酶(MTHFR)基因的分布.方法 选择高血压患者共998例进行MTHFR基因检测.方法 采用聚合酶链式反应-限制性片段长度多态性(PCR-RFLP)法,精准定位基因型;应用高效液相色谱荧光检测法测定血浆同型半胱氨酸(HCY).结果 参与检测的高血压患者共998例(男494例,女504例),MTHFR基因检测结果 为TT型82例(8.22%),CT型338例(33.87%),CC型578例(57.92%).其中男性TT型54例(10.93%),CT型168例(34.01%),CC型272例(55.06%),女性TT型28例(5.56%),CT型170例(33.73%),CC型306例(60.71%).男性TT型比例高于女性(10.93%vs 5.56%,P<0.01).同时进行检测HCY者共有739例(男351例,女388例),为H型高血压者389例(52.64%),男性224例(63.82%),女性165例(42.53%),男性比例也高于女性.结论 佛山地区TT型为8.22%,基因频率低.H型高血压为52.64%,偏低.表明基因多态性分布存在地区差异.H型高血压及TT型基因男性均高于女性.

A Comparative Analysis of Methylene Tetrahydrofolate Reductase Gene Detection Results between Foshan and Nationwide Results

Objective To investigate the Methylene tetrahydrofolate reductase ( MTHFR ) gene distribution a-mong hypertensive patients in Foshan and compare with nationwide results .Methods MTHFR gene detection was per-formed in 998 cardiovascular outpatients and hypertensive inpatients in the Second People ′s Hospital of Foshan from April 2013 to September 2014.The polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) method was applied for accurate gene mapping .Plasma HCY was determined by high -performance liquid chromatography with fluorescence detection (HPLC-FLD).Results Among 998 hypertensive patients (494 males and 504 females) selected for MTHFR gene detection , 82 patients were detected to be TT ( accounting for 8.22%) , while 338 patients were identi-fied as CT (accounting for 33.87%) and 578 patients as CC (accounting for 57.92%).Among them, there were 54 TT (10.93%) and 168 CT (34.01%) in males, as well as 28 TT (5.56%) and 170 CT (33.73%) in females.For TT type, the proportion of males was significantly higher than that of females (0.1093 vs.0.0556, P<0.01).Meanwhile, 739 patients were chosen for HCY detection (351 males and 388 females), and there were 389 H type hypertensive pa-tients, accounting for 52.64%, among whom there were 224 males (63.82%) and 165 females (42.53%).The propor-tion of males was significantly higher than that of females .Conclusion The TT gene type frequency in South China ( Fos-han) (8.22%) is lower than 25%, which is the nationwide frequency .The H type hypertension frequency in South China (52.64%) is also lower than 75%, which is the nationwide frequency .It is shown that there are regional differences in the distribution of gene polymorphism .The frequency of H type hypertension and TT type gene in males are higher than those in females .
