马勃多糖CGP-Ⅱ抗S180肉瘤及体外抗氧化研究

摘    要:目的:探讨马勃酸性多糖CGP‐Ⅱ抗S180肉瘤的作用及体外抗氧化作用。方法:水提醇沉法制得马勃粗多糖,再经 HiTrap Capto DEAE阴离子交换层析和Sephacryl S‐300 HR凝胶柱层析进一步纯化得到马勃酸性多糖CGP‐Ⅱ。采用咔唑‐硫酸法测得糖醛酸含量,同时建立小鼠S180肉瘤移植模型,研究CGP‐Ⅱ的体内抑瘤效应。利用氧自由基吸收法(Oxygen Radical Absorption Capacity ,ORAC)、DPPH法评价其体外抗氧化能力。结果:CGP‐Ⅱ糖醛酸含量为17.2%,200 mg/(kg?d)剂量组多糖对S180肉瘤的抑瘤率为48.4%,CGP‐Ⅱ具有一定的DPPH 自由基清除能力和氧自由基吸收能力。结论:酸性多糖CG P‐Ⅱ具有一定的抗肿瘤活性及体外抗氧化作用,本研究为其进一步开发利用奠定了基础。

Lsolation,Purification of Acidic Polysaccharide from Calvatia Gigantea and Its Anti-S180 Sarcoma Activity
Abstract:Objective :To study the anti‐S180 activity and antioxidant effects of acidic polysaccharide from Calvatia gigantea .Methods:Water extracting and ethanol precipitating methods were applied to extract polysaccharide from Calvatia gigantea .Mice tumor model was established with transplantion of sarcoma S 180 .In addition ,antioxidant activity of CGP‐Ⅱwas evaluated by DPPH free radical scavenging assay and oxygen radical absorbance capacity (ORAC) assay .Results :The content of uronic acid in CGP‐Ⅱ was 17.2% .The inhibition rates of CGP‐Ⅱ on sarcoma S180 in mice was 48.4% ,when the dosage of CGP‐Ⅱ reached to 200 mg/(kg ? d) .CGP‐Ⅱ showed antioxidant capacity against DPPH and oxygen radical absorption capacity .Conclusion:CGP‐Ⅱ showed obvious antitumor and antioxidant effects .This research will provide important basis for the further research and application of CGP‐Ⅱ .
