脑灵汤对Alzheimer病模型大鼠行为学及海马胆碱能神经系统的影响

摘    要:目的观察脑灵汤对阿尔茨海默病(AD)模型大鼠学习记忆能力,以及对海马部位乙酰胆碱酯酶(AChE)和胆碱乙酰基转移酶(ChAT)的影响。方法D-半乳糖腹腔注射联合三氯化铝(AlCl3)皮下注射诱导建立大鼠AD模型,造模前后进行Y-电迷宫实验检测各组大鼠学习记忆能力,药物干预后检测各组大鼠学习记忆能力、海马区AChE及ChAT活力。结果治疗30 d后脑灵汤组学习记忆成绩比西药组及模型组显著提高(P均<0.05),海马区AChE活力较模型组及西药组显著降低(P均<0.01),ChAT活力较模型组及西药组显著升高(P均<0.01)。结论脑灵汤能降低大鼠海马组织区乙酰胆碱酯酶活力,升高胆碱乙酰基转移酶活力,改善脑内胆碱能系统的功能,从而提高AD模型大鼠学习记忆能力。

Influence of Naoling decoction on praxiology and cholinergic nerve system in Alzheimer's disease model rat
Abstract:Objective It is to observe the influence of Naoling decoction(NLD) on learning and memory,and the activity of Acetylcholinesterase(AChE) and choline acetyl transferase(ChAT) in Hippocampus in the Alzheimer's disease(AD) model rat.Methods The AD model of rat was established by peritoneal injected D-galactose and subcutaneous injected AlCl3.The learning and memory was observed by Y-electric maze experiment before and after model made,and post-treatment.The activity of AChE and ChaT of AD model rats were measured after treated for 30 days. Results The learning and memory in NLD group were increased more than that in model group and Western medicine group significantly(P<0.05).The activity of AChE in NLD group was less than that in Western medicine group and model group significantly(P<0.01).The activity of ChAT in NLD group was increased more than that in model group and Western medicine group significantly(P<0.01). Conclusion NLD can decrease the activity of AChE and enhance the activity of ChAT in hippocampus of model rat,which may be the mechanisms of improving the learning and memory of AD rat
