摘    要:目的观察并研究盐酸阿米替林在大鼠体内死亡后的分布。方法建立盐酸阿米替林的中毒致死大鼠模型,大鼠经2倍LD50(39.2mg/kg)剂量盐酸阿米替林灌胃后,观察中毒症状,死亡后取脑、心、肺、肝、脾、肾、肌肉(左腿)、心血、周围血等组织、脏器和体液,薄层色谱扫描法检测其中盐酸阿米替林含量。结果大鼠在灌药后3~5min即出现抽搐症状,间隔3min抽搐1次,每次抽搐持续2~4秒,2h内死亡。2倍LD50剂量盐酸阿米替林灌胃后,各脏器阿米替林浓度由高向低分别为肺〉肝、肌肉、肾、脾〉心、脑、心血、外周血。结论使用薄层色谱分析法检测盐酸阿米替林,其方法简便、快速,中毒死亡大鼠肺、肝、肾、脾和肌肉中盐酸阿米替林含量较高,可作为疑似阿米替林中毒案件毒物分析的检材。

Abstract:Objective To observe and study in vivo postmortem distribution of amitriptyline hydrochloride in rats. Methods Poisoned rat models were set up with amitriptyline hydrochloride. After gastric infusion of amitriptyline hydrochloride of 2-time LDs0 (39.2 mg/kg),the poisoning symptoms were observed. After death,the tissues,organs and body fluids were collected,such as brain,heart,lung,liver, spleen, kidney, muscle (left leg), cardiac blood, peripheral blood, etc. Thin layer chromatography scanning was used to detect the concentration of amitriptyline hydrochloride in each specimen. Results Spasm and convulsion presented in 3-5 rain after infusion in rats, one attack every 3 min,lasting 2-4 sec in each attack. The rats were died in 2 h. After gastric infusion of amitriptyline hydrochloride with 2-time LDs0 dose,it was discovered that the concentrations of amitriptyline hydrochloride in every organ were different,named lung〉liver-muscle-kidney-spleen〉heart-brain-cardiac blood-peripheral blood. Conclusion Thin layer chromatography scanning is convenient,sample and rapid in detection of amitriptyline hydrochloride concen- tration. The concentration of it is quite higher in lung,liver,kidney and spleen as well as in muscle of died rats due to poisoning. Hence, the materials may be applied to the analysis of suspended cases of amitriptyline poisoning.
