柴胡挥发油对大鼠肝能量代谢的影响

摘    要:目的研究柴胡挥发油致大鼠肝毒性损伤的能量机制。方法大鼠ig给予柴胡挥发油0.19,0.28和0.42 m·lkg-1,连续15 d。Clark氧电极法测定肝线粒体呼吸耗氧量、呼吸控制率(RCR)和磷氧比值(P/O),测定ATP含量及定磷法测定ATP酶活性的变化。结果与正常对照组比较,柴胡挥发油0.42ml·kg-1可使大鼠肝线粒体和P/O从(1.76±0.28)%和1.28±0.38分别降至(0.76±0.12)%和0.71±0.21,呼吸耗氧量从(2.82±0.64)mol·min-1·g-1降至(1.04±0.23)mol·min-1·g-1,ATP含量从(1.00±0.18)μmol.g-1蛋白降至(0.72±0.13)μmol.g-1蛋白,Na+-K+-ATP酶、Mg2+-ATP酶和Ca2+-ATP酶活性分别从0.94±0.14,0.95±0.14和(0.97±0.16)mmol·h-1·g-1降至0.53±0.08,0.67±0.10和(0.65±0.11)mmol·h-1·g-1。柴胡挥发油0.28和0.19 ml·kg-1使大鼠肝脏线粒体PCR、P/O、呼吸耗氧量、ATP含量和ATP酶活性亦均显著降低(P<0.01)。结论柴胡挥发油可通过抑制线粒体呼吸功能、影响肝脏能量代谢造成肝毒性损伤。

Abstract:OBJECTIVE To explore the energy mechanism of volatile oil from Bupleurum chinese for hepatotoxic injury. METHODS Volatile oil from B. chinese 0.19, 0.28 and 0.42 ml·kg-1 was ig given to rats for 15 d continually. The changes in respiratory function, contents of ATP and acitivities of ATPases of hepatic mitochondrion were detected. RESULTS Compared with normal control group,the respiratory control rate (RCR) and phosphorus/oxygen rate(P/O) reduced from(1.76±0.28)% and 1.28±0.38 to (0.76±0.12)% and 0.71±0.21 in volatile oil from B. chinese 0.42 ml·kg-1 group, respectively, and the respiratory oxygen uptake was reduced from (2.82±0.64) to (1.04±0.23)mol·min-1·g-1. Contents of ATP were reduced from (1.00±0.18) to (0.72±0.13)μmol·g-1, and activities of Na+-K+-ATPase, Mg2+-ATPase and Ca2+-ATPase from 0.94±0.14, 0.95±0.14 and (0.97±0.16)mmol·h-1·g-1 protein to 0.53±0.08, 0.67±0.10 and (0.65±0.11)mmol·h-1·g-1 protein, respectively. Compared with normal control group, RCR, P/O, respiratory oxygen uptake, contents of ATP and ATPases in in volatile oil from B. chinese 0.28 and 0.19 ml·kg-1 groups were also significantly reduced. CONCLUSION Volatile oil from B. chinese can cause hepatotoxicity in rats by inhibiting function of hepatic mitochondrion and influence the energy metabolism of the liver.
