摘    要:目的:探讨在重组合异种骨(RBX)移植局部白细胞介素—8(IL-8)mRNA和蛋白质的表达规律.方法:将重组人骨形成蛋白(rhBMP2)复合于牛松质骨中制成RBX并植入小鼠股部肌肉内,于术后4,7,14和21d取材,应用原位杂交和免疫组织化学技术检测植骨局部IL-8 mRNA及蛋白的表达与细胞定位.标准对照组动物植入单纯牛松质骨,并另设空白对照.结果:术后4,7,14和21d,实验组和标难对照组动物植骨局部均可检测到IL—8mRNA及蛋白的表达.叫主要定位于单核细胞、巨噬细胞、中性粒细胞和成纤维细胞以及部分软骨细胞、成骨细胞和新生骨髓中部分骨髓细胞.结论:在RBX移植局部多种细胞表达IL—8,提示RBX可能通过新生骨中的多种细胞成分表达IL—8,参与异种骨移植的局部免疫反应。

Abstract:AIM: To observe the expression of interleukin 8 (IL 8) mRNA and protein in the area of implanted reconstituted bone xenograft. METHODS: The reconstituted bone xenografts which combined the bovine cancellous bone particles and recombined human bone morphogenic protein 2 (rhBMP2) were first implanted into the muscle of mice and then were removed 4, 7, 14 and 21 days later. IL 8 mRNA and protein in the site of implant were assayed by in situ hybridization and immunohistochemistry. In the standard control group, cancellous bone particles with no rhBMP2 were implanted. The blank control group was also set up and observed. RESULTS: The expression of IL 8 mRNA and protein was observed in the area of implantation at 4, 7, 14 and 21 days after implantation. IL 8 was expressed mainly by monocytes, macrophages, fibroblasts, chondrocytes, osteoblasts and marrow cells of new formed marrow. CONCLUSION: Many different kinds of cells express IL 8 mRNA and protein in the area of reconstituted bone xenograft, which may suggest that the reconstituted bone xenograft plays a role in the local immunity of bone transplantation through multiple types of cells that express IL 8.
