摘    要:目的:探索水苏糖酸水解制备甘露三糖的方法。方法:通过HPLC定量检测,采用正交试验设计,考察温度、pH值、时间三个影响因素,来确定水苏糖的酸水解工艺;并经柱层析分离得到甘露三糖。结果:在本实验条件下,温度和时间对水苏糖酸水解影响不显著(P<0.05),而酸度对其有显著性影响(P>0.05),确定最佳反应条件为90℃、24 h、pH 2.5。结论:水苏糖酸水解制备甘露三糖的方法可行。

Abstract:OBJECTIVE: To optimize the processing of mannatriose preparation from stachyose by acidolysis. METHODS: An orthogonal experiment was performed to select a better condition of acidolysis. Fructoes and stachyose was determined by HPLC. RESULTS: Among the factors set in the present work, temperature and time have little influence on the acidolysis, while the effect of pH value was significant (P < 0.05). The best processing was selected as heating stachosy solution for 12h at pH 2.5 and 90 degrees C. CONCLUSION: It is practicable to prepare manninotriose from stachyoses by acidolysis.
