摘    要:目的建立水牛角浓缩粉中6种氨基酸成分的含量测定方法和特征图谱分析方法,分析不同厂家生产水牛角浓缩粉及其易混品质量差异。方法采用异硫氰酸苯酯(PITC)柱前衍生化法建立水牛角浓缩粉氨基酸成分含量分析方法和总氨基酸HPLC特征图谱。结果建立的水牛角浓缩粉特征图谱分析方法稳定可行,可同时测定6种氨基酸成分含量,测得14个氨基酸类特征峰;测得12批次水牛角浓缩粉分为高和低两个明显的质量差异组,其中高含量组与羚羊角样品氨基酸成分含量相近,但有3个特征峰相对含量差异较大。结论建立的水牛角浓缩粉总氨基酸特征图谱分析方法可用于水牛角与易混品羚羊角的区别鉴定,为水牛角浓缩粉的质量控制提供了研究基础。

Abstract:Objective To establish a simultaneous determination of six amino acids and specific HPLC characteristic chromatogram method for the quality evaluation of powerdered buffalo horn extract, and to analysis quality difference of Powerdered Buffalo Horn Extract and its confusing products. Methods Use RPHPLC method with phenyl isothiocyanate(PITC) derived after derivatization reagent to establish characteristic chromatogram of total amino acids in powerdered buffalo horn extract. Results The method can be used to detect6 amino acids, and 14 characteristic peaks of amino acids are obtained. 12 batches of powerdered buffalo horn extract are divided into high quality group and low quality group. Amino acid composition and content of antelope horn is similar to the high quality group, but three characteristic peaks are obviously different. Conclusion The characteristic chromatogram of total amino acids can be used to control the quality of powerdered buffalo horn extract, and can distinguish antelope horn.
