虎杖苷通过激活Nrf2/HO-1信号通路减轻大鼠急性心肌梗死后心肌细胞损伤

摘    要:目的研究虎杖苷对大鼠急性心肌梗死(AMI)后心肌细胞损伤的保护作用及分子机制。方法 SD雄性大鼠随机分为Sham组、AMI组、虎杖苷组、虎杖苷+ZnPP组。后3组通过左冠状动脉前降支结扎的方式建立AMI模型。虎杖苷为每天40 mg/kg灌胃,血红素加氧酶1(HO-1)抑制剂Zn PP为每天20 mg/kg腹腔注射。造模后第8天,检测各组心功能超声指标左心室射血分数(LVEF)、左室内压力变化最大上升和下降速率(±dp/dtmax)、血清心肌损伤指标、心肌梗死面积、心肌氧化应激指标及核因子E2相关因子2(Nrf2)/HO-1表达情况。结果与Sham组比较,AMI组LVEF、+dp/dtmax、-dp/dtmax及心肌超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的含量均明显降低,心肌梗死面积及血清乳酸脱氢酶(LDH)、磷酸肌酸激酶(CK)、磷酸肌酸激酶同工酶(CK-MB)含量及心肌丙二醛(MDA)的含量及Nrf-2、HO-1的表达水平均明显增加(P0. 05)。与AMI组比较,虎杖苷组LVEF、+dp/dtmax、-dp/dtmax及心肌SOD、GSH-Px的含量及Nrf-2、HO-1的表达水平均明显升高,心肌梗死面积及血清LDH、CK、CK-MB含量及心肌MDA的含量均明显降低(P0. 05)。与虎杖苷组比较,虎杖苷+ZnPP组LVEF、+dp/dtmax、-dp/dtmax及心肌SOD、GSH-Px的含量均明显降低,心肌梗死面积及血清LDH、CK、CK-MB含量及心肌MDA的含量均明显增加(P0. 05)。结论虎杖苷能够减轻大鼠急性心肌梗死后心肌细胞损伤,该效应可能由Nrf2/HO-1通路的激活介导。
