苯甲酸雌二醇对去卵巢脑缺血大鼠NOS及AChE的影响

摘    要:目的探讨苯甲酸雌二醇对去卵巢脑缺血大鼠的脑保护作用及其可能机制。方法采用改良4-VO方法将去卵巢雌性大鼠制备成脑缺血模型,应用水迷宫检测大鼠学习记忆能力,取脑组织测定一氧化氮合酶(NOS)及乙酰胆碱酯酶(AChE)的活力变化,同时观察海马区病理学改变。结果与假手术组比较,缺血组海马区神经元损伤明显,学习记忆能力明显减退(P〈0.01),脑组织中NOS活力明显下降(P〈0.01),AChE的活力明显增加(P〈0.01);与缺血组比较,治疗组海马区神经元损伤较轻,学习记忆能力有明显改善(P〈0.01),NOS活力增强(P〈0.05),AChE活力有所降低(P〈0.05)。结论苯甲酸雌二醇可以减轻因去卵巢脑缺血所致的大鼠认知功能障碍,可能与改变脑组织中NOS和AChE的活性有关。

Abstract:Objective: To study the neuroprotection of estradiol benzoate on ovariectomized rats of cerebral ischemia and its mechanism of action.Methods: The cerebral ischemia model was made by reforming 4-VO method(pairs of side vertebra artery electricity congeal and pairs of side common carotid artery insert closing),the change of learning and memory function was tested using Morris water maze,the levels of NOS and AChE in hippocampus were determined,and the changes of morphology in hippocampal CA1 were observed with microscopy.Results: Compared with sham group,the pathological impairment of the hippocampal neuron showed obvious ingravescence,the activity of NOS were reduced(P<0.01)and the activity of AChE were increased,(P<0.01),and the ability of learning and memory were decreased in the ischemia group(P<0.05);Compared with those of ischemia group,the above mentioned indices of the treated group changed obviously,and there was significant difference(P<0.01 or P<0.05).Conclusion: Estradiol benzoate can improve cognitive dysfunction induced by cerebral ischemia in ovariectomized rats,which may be related to the levels of NOS and AChE in brain.
