腺病毒载体介导的单纯疱疹病毒-胸苷激酶基因杀伤人膀胱癌细胞的实验研究

摘    要:目的探讨HSV-tk/GCV系统基因治疗膀胱癌细胞的方法及结果。方法与结果采用了带有CMV启动子驱动HSV-tk基因的重组腺病毒载体,体外转染HTB9细胞后对GCV敏感性增强。当MOI=100时,HTB9细胞的IC50为2.5mg/L,病毒浓度降低,GCV对细胞的杀伤效率亦降低。将不同比例的tk 细胞和tk-细胞混合后加入10mg/L GCV,发现HSV-tk/GCV系统有很强的“旁观者效应”。结论HSV-tk/GCV系统是一种有效、安全地治疗膀胱癌的新方法。

Abstract:Objective To study the new way of adenovirus-mediated gene therapy for bladder cancer.Methods and Results We used the replication-defective adenovirus carring the HSV-tk gene(Ad.tk) under control of the cytomegalovirus(CMV).In vitro experiments,the HTB9 cells transferred by tk gene were killed in the presence of GCV treatment.When tk cells were mixed with their parental cells at various ratios,GCV not only killed tk cells,but neighboring cells without tk gene also killed by bystander effect.Conclusion Adenovirus-mediated HSV-tk/ GCV system may be used as apromising suicide gene therapy for bladder carcinoma.
